辣椒不同外植体离体培养再生

【摘要】 8个品种的辣椒子叶和下胚轴接种在附加不同植物激素的MS培养基上,经过芽诱导、芽伸长、生根、移栽入盆四个阶段,得到完整的再生植株。实验比较了苗龄、不同的外植体类型对分化率的影响,结果发现8~10天的苗是最佳的,子叶的分化率高于下胚轴。培养基中添加AgNO3明显促进外植体的分化,GA3是芽伸长的关键因子,IBA的生根效果要好于无激素MS培养基。通过这一过程,建立起一个基于最佳激素组合的高效植株再生体系。

【Abstract】 Regenerated plants were obtained from cotyledon and hypocotyl explants ofeight pepper varieties(Capsicum annuum L.)cultured on basal MS medium supplementedwith different kinds and different concentrations of plant hormone by four--step cultureprocedure including shoot induction, shoot elongation, rooting of excised shoot andtransplanting into soil. The effect of seedling stage on frequency of shoot induction wasobserved with a result that seedlings of 8~10 days old were best for shoot induction. Ingeneral, the frequency of shoot induction from cotyledon explants was higher than thatfrom hypocotyl explants in terms of explant types. AgNO3 significantly increased thefrequency of shoot induction from all varieties. GA3 was the key factor in shootelongation. The elongated shoots cultured on MS basal medium were easier to root thanthat on MS basal medium with IBA. Through above four--step procedure, a genetictransformation system has been established.
