柑橘细胞工程研究进展

【摘要】 本实验室自上一世纪80年代以来一致开展柑橘细胞工程研究,在柑橘愈伤组织诱导、原生质体分离、培养和融合、体细胞杂种鉴定、体细胞杂种评价利用等方面开展了大量的工作,取得了一定的成就。诱导了20余个基因型的胚性愈伤组织,有6个基因型原生质体培养再生植株,采用PEG和电场诱导融合获得了20余例柑橘体细胞杂种,体细胞杂种的鉴定由形态学、细胞学和分子标记三个方面构成。体细胞杂种的应用集中在两个方面:第一,作为砧木,现有研究表明体细胞杂种的抗寒性、抗热性和抗脚腐病主要介于双亲之间;第二,作为倍性育种亲本,将四倍体体细胞杂种与二倍体单胚或单多胚类型杂交,结合幼胚培养获得了近10个组个的三倍体植株。体细胞杂种的核质分析表明。
【Abstract】 since 1980s much effort has been directed to citrus cell engineering,involving induction of callus, isolation, culture and fusion of protoplasts, identification ofsomatic hybrids, evaluation of agronomic interest of the somatic hybrids. To dateembryogenic calluses have been successfully induced from more than 20 genotypes. Plantswere regenerated from protoplasts of 6 genotypes. In addition more than 20 intergenericand interspecific somatic hybrids have been obtained from PEG or electricity--mediatedfusion. Identification of the somatic hybrids were based on cytology, morphology andmolecular markers. For the time being the somatic hybrids are evaluated for their potentialas rootstock. The results available showed that the somatic hybrids were in between thefusion parents concerning the resistance to cold, hot and foot rot. On the other hand thesomatic hybrids are used as breeding parent for interploid crosses between tetraploidsomatic hybrids and monoembryonic or poly and monoembryonic cultivars, from whichtriploid plants have been produced from nearly 10 combinations.
