小麦幼穗离体培养高效再生系统的初步研究

【摘要】 针对小麦组织培养中基因型障碍和植株再生频率偏低等问题,选择5个不同小麦基因型的不同时期的幼穗为外植体,研究对离体培养的反应与再生效果。结果表明,虽然不同基因型最适离体培养时期有所差异,一般与愈伤组织的生长速度有一定的关系,但总体上具有明显的一致性趋势,即幼穗最适取材时期在护颖原基形成期至雌雄蕊原基形成期之间。研究发现产生变态苗、绿点(有根无苗)的愈伤组织在分化培养之后增加一个芽苗的诱导及伸长阶段能有助于继续分化成苗。绿苗形成之后,进行促根壮苗培养,同时加强试管苗移栽后的管理,有效地提高了试管苗移栽的成活率,在此基础上建立了一套高效的小麦幼穗组培再生系统。
【Abstract】 There have been problems of genotype dependence and low frequency ofplant regeneration in wheat tissue culture. In order to solve these problems, 5 genotypesof wheat were chosen to study the reaction and regeneration effect of in vitro culture.Young spikes at different stages were used as explants. Difference in optimum stage ofyoung spike culture existed in different genotypes, which related to the growth speed ofcallus to a certain extent. But in general there was still an obvious same tendency thatoptimum sampling time for young spike culture was between the formation stage ofprotective glume primordium and formation stage of pistil and stamen primordium. Tothose calli on which abnormal plants and green points (with roots but without leaves)generated, adding a culture process of green plant induction and elongation after thedifferentiation culture could help them keep on differentiating and develop complete plantsat last. The survival rate of transplanting could be increased effectively by conducting aculture procedure to promote root development and to make the plants grow stronger afterthe formation of green plants and then tightening up the management after the in vitrocultured plants were transplanted. An efficient regeneration system for young spikeculture of wheat has been set up on the basis of such methods.
