基于叶绿体trnL-trnF基因间区的楸树属部分种植物的分子系统学研究

【摘要】 设计楸树trnL-trnF基因间隔区序列的特异引物,扩增并分析了楸树属16个种/类型 trnL-trnF基因间隔区序列,其长度在710bp和939bp之间,且富含A/T,比较长度为659bp。该序列一共具有34个分析性状(包括17个变异位点,9个位点碱基缺失)。用DNAMAN软件对16份材料的 trnL-trnF基因间隔区序列进行聚类分析,建立了系统进化发育树。研究表明利用trnL-trnF基因间隔区序列来进行楸树属内树种的系统分类分析支持形态学上关于大类别的划分方法,但把光叶楸从楸树组中分开,归为灰楸组。该研究结果为楸树属树种全部类型的分类提供了参考。

【Abstract】 Special primer for the trnL-trnF introns was designed. The sequences of segments of the trnL-trnF introns from 16 species of Catalpa Scop, were cloned and analyzed. Sequence length varied from 710bp to 935bp, and base composition was with high A+T, of which 659bp was compared. There were 34 analyzing characters could be used for analysis, including 17 transpositional loci and 9 missing ones. The strict consenus trees were built useing DNAMAN software. The cpDNA results on Catalpa Scop’s classification were consistent with that of morphology except that guangyeqiu wasn’t belong to C. bungei C.A. Meyer but C. fargesii Bureau. This study helped to build the whole tree system of Catalpa.
