ISSR标记的精度、适用范围及应用开发新策略——以牡丹分析为例

【摘要】 牡丹为芍药科(Paeoniaceae Rudolphi.)芍药属(Paeonia L.)牡丹组(Sect.Moutan)落叶灌木,是重要的观赏绿化植物和资源植物,在中国已有2 000余年的栽培历史,目前约有1 500个中国牡丹观赏品种。牡丹品种通常根据花型和花色进行品种分类。目前主要存在三大不同的品种分类体系[①喻衡(1998)认为存在11种花型,8大色类,②王莲英(1997;1999)提出存在16种花型,8大色类,以及③李嘉珏(1999)认为存在14种花型,10大类花色],争议较大。对刁二“多花型品种”或间于上述各种花型的“过渡性花型”等问题,现有的品种分类体系都不能给出令人满意的解决方案。在牡丹各产区,品种来源记载不详,存在同物异名、异物同名现象,品种鉴定困难严重制约着中国牡丹产业的发展(陈富慧等,2005;洪德元和潘开玉,1999;李嘉珏,1999;王莲英,1997;喻衡,1998)。

【Abstract】 Tree peonies are deciduous shrubs belonging to genus Paeonia L., section Moutan DC., family Paeoniaceae Rudolphi. They are important ornamental and resource plants, having-been cultivated in China for more than 2000 years, currently there are about 1 500 cultivars. Chinese tree peony cultivars usually are classified based on characters of flower form and color. At present, there mainly exist three different classification systems of Chinese tree peony cultivars, which are under dispute. Yu (1998) reported 11 flower forms and 8 categories of colors, Wang (1997; 1999) reported 16 flower forms and 8 categories of colors, whereas Li (1999) believed there are 14 flower forms and 10 categories of colors. However, current cultivar classification systems could not provide satisfactory solutions concerning those plants with multiple flower forms or transitional flower forms. In tree peony production areas, development of tree peony industry has been seriously limited, because of the difficulty in cultivar identification caused by unclear documentation of origin of the cultivar and existence of synonyms and homonyms (Chen et al., 2005; Hong and Pan, 1999; Li, 2005; Wang, 1997; 1999; Yu, 1998). Since morphological characters are affected by environmental influences and developmental levels, the amount of morphological markers for cultivar classification is limited, it is necessary to further study and develop a more effective classification and identification method at DNA molecular level. Since no ideal results were obtained in screenings of the published arbitrary ISSR primers available, we began to design new ISSR primers through collocation and recombination of sequences of the arbitrary ISSR primers which had produced bands in PCR amplifications. Some good ISSR primers were obtained by screenings of the newly designed ISSR primers. Cloned sequencing of a 695bp ISSR band shared by 3 tree peony cultivars indicated that there existed 1 to 3 variable single-nucleotide-site (s) among sequences of different clones of the DNA band within cultivar, and 2 to 5 variable single-nucleotide-sites in sequence of the ISSR band a-mong cultivars. However, these single-nucleotide-site variations could not be used for cultivar classification, because they existed both within cultivar and among cultivars. As a result, an ISSR electrophoretic band, in fact, is a mixture of different types of ISSR fragments which are equal in length, highly similar and very slightly different in sequence. Thus, strictly speaking, the ISSR band shared among cultivars could be only considered as an identical molecular character shared among cultivars at ISSR marker scale, the ISSR fragments in different length (polymorphic bands) obtained simultaneously using the same primer among cultivars could be regarded as conclusive evidences of the difference among cultivars, polymorphic band data can be used to reveal genetic relationships among tree peony cultivars.
