两种新型聚氨酯涂层材料的血液相容性研究

【摘要】 以十八烷基聚氧乙烯(SPEO)与4,4’-二苯甲撑二异氰酸酯(MDI)的偶联物SPEO-MDI-SPEO(MSPEO)作为改性材料,通过血小板黏附实验,复钙化时间和凝血酶原时间的测定以及动态抗凝血测试,对比了以聚醚氨酯(PEU)与壳聚糖(Chi)为成膜剂时对提高聚氨酯基材抗凝血性能的作用。结果发现以PEU和壳聚糖为成膜剂的两种表面涂层均可显著提高聚氨酯材料的抗凝血效果,并不引发溶血反应,是极具潜力的两种新型医用聚氨酯涂层材料。其中,壳聚糖为成膜剂的表面虽然对血小板的吸附较PEU为成膜剂的表面要多,但能更有效延长复钙化时间,对外源性凝血系统阻抗效果较PEU为成膜剂的改性体系要好,而后者则能有效延长凝血酶原时间,对内源性凝血系统的阻抗效果较好。
【Abstract】 Amphiphilic coupling-polymer of stearyl poly (ethylene oxide )-co-4, 4’-methylendiphenyl diiso-cyanate-co- stearyl poly (ethylene oxide), for short MSPEO, was specically designed as surface-modifying additives. The blends of MSPEO in both polyurethane (PEU) and chitosan(Chi) were investigated as the coating materials for intravascular device. Two kinds of static clotting time tests, PRT and PT, as well as the static platelet adhesion experiment were carried out. And the dynamic experiment was performed with a closed-loop tubular system under a blood shear rate of 1500 s-1.The results showed that both blend coatings can improve the anti-coagulation of polyurethane greatly and will not lead to hemolysis. Although more platelets were adhered to the surface modified by Chi-MSPEO blend coating than to the surface modified by PEU-MSPEO blend coating, the surface modified by Chi-MSPEO has longer plasma recalcification time. But the surface modified by PEU-MSPEO has longer prothrombin time. So the surface modified by Chi-MSPEO has better anti-coagulation in the exogenetic blood-coagulation system,while the surface modified by PEU-MSPEO has better anti-coagulation in the endogenet-ic blood-coagulation system.
