猪卵母细胞体外成熟培养时间及去核方法的研究

【摘要】 对不同体外成熟培养时间的猪卵母细胞进行盲吸法去核及利用脱羧秋水仙碱(DC)和放线菌酮(CHX) 的化学诱导法去核效率进行了比较研究.用盲吸法去核,体外成熟培养(IVM)39~41 h 组卵母细胞去核率与 IVM 36~38 h 组差异显著(P<0.05),与42~44 h 组差异不明显(P>0.05),但3组显著高于 IVM 45~48 h 组 (P<0.05).化学诱导法去核结果,IVM 42~44 h 组去核率最高,但与 IVM 39~41 h 组差异不显著(P>0.05), 其他各组间均差异显著(P<0.05).在相同的体外成熟培养时间内,盲吸法的去核率比化学诱导法的去核率要高.猪卵母细胞 IVM 在39~44 h 内去核率较高,且盲吸法去核效率高于化学诱导去核.
【Abstract】 This experiment was conducted to study enucleated rate of porcine oocytes cultivated with different in vitro mat- uration times,which was treated with physical enucleation and chemical-induced enucleation involved in Demecolcine and CHX.The results show that when maturation time were 39-41 h,the enucleated rate with physical enucleation method was significantly different from that in IVM 36-38 h(P<0.05),while no significant difference from that in IVM 42- 44 h(P>0.05),and that in all three groups was higher than that in IVM 45-48 h(P<0.05).Whereas when in vitro maturation time were 42-44 h,the enucleated rate with chemical-induced enucleation was the highest,which was in- significantly different from that in IVM 39-41 h(P>0.05)and there was significant difference between the other groups(P<0.05).With the same in vitro maturation time,the rate by physical enucleation was higher than that resulted in chemical-induced method.The study indicated that the higher enucleation rate of porcine oocytes can be achieved with IVM 39-44 h and the efficiency of physical enucleation can better than that of chemical-induced method.
