棘阿米巴致细胞程序化死亡的初步研究

【摘要】 由佩戴隐形眼镜而引起角膜炎患者的角膜切片中分离到一株棘阿米巴,在体外用此棘阿米巴的裂解液能诱导肿瘤细胞发生程序化死亡或凋亡。用流式细胞仪定量测定了凋亡细胞的百分数,琼脂糖凝胶电泳显示了凋亡细胞出现的典型的180-200bp碱基对递减的规律性DNA梯度片段。而未用阿米巴裂解液处理的肿瘤细胞则不发生这些现象。此结果提示棘阿米巴能诱导细胞凋亡。可能是此棘阿米巴主要的溶细胞机理。
【Abstract】 Tumor cells undergoing apoptosis were induced by the Acanthamoeba isolated from keratitis patients wearing soft-contact lens. Typical ladders pattern of discontinuous DNA fragments consisting of multimers of 180- 200bp were formed during agarose gel electropharesis, when tumor cells were exposed to Acanthamoeba extracts. The number of tumor cells producing apoptosis increased significantly in a dose-dependent fashion following content of the parasite extract while detected by flow Cytometry. By contrast, similar effects were not found in tumor cells exposed to non-acanthamoeba extracts. The results suggest that the Acanthamoeba is able to lyse tumor cells by a mechanism of apoptosis.
