PEG修饰大蒜素长循环脂质体的工艺优化
Optimization of the formula and technique for allitridin long-system liposomes modified by PEG

【摘要】 目的:制备PEG修饰大蒜素长循环脂质体(PEG-DATS-LCL)。方法:采用Szoka改进逆相蒸发技术,制备PEG修饰大蒜素长循环脂质体,以高效液相色谱法测定大蒜素(DATS)的包封率和载药量,用均匀设计法优化脂质体处方和制备工艺。结果:最佳工艺条件为:胆固醇与卵磷脂量的比(Chol:EPC)为0.8:1、大蒜素与卵磷脂量的比(DATS:EPC)为1:4、缓冲溶液PH值为6.8、有机溶媒蒸发温度(T)为30℃。结论:制得的脂质体粒子圆整,分布较均匀,算术平均粒径为12.63 μ m,平均包封率为(90.77±2.14)%;载药量为(15.11±1.67)%。
【Abstract】 OBJECTIVE To prepare allitridin long-system liposomes modified by PEG. MATHODS PEG-DATS-LCLs were prepared by Szoka converse vaporized mathod. The uniform design coupled with computerized optimization was adopted to screen the formulation and preparation process. A method to determine Allitridin in rabbit plasma by high performance liguid chromatography (HPLC) was established. RESULTS (Chol: EPC) was 0.8 : 1, (DATS: EPC) was 1 : 4, pH was 6.8, Temperature was 30℃.CONCLUSION Liposomes were globular in appearance and dispersed well. The mean diameter was 12.63um, the entrapment efficiency is 90.77% and loading capacity was 15.11%.
