【摘要】 目的:研究γ-干扰素(γ-IFN)对他莫昔芬(TAM)抗乳腺癌细胞的增强作用。方法:在体外培养条件下,分别或联合应用γ-干扰素、TAM或雌二醇(E2)作用于ER阳性的MCF-7和 ER阴性的MDA-MB-231人乳腺癌细胞株,用MTT比色法分析细胞生长抑制作用,流式细胞仪检测细胞周期分布,DNA凝胶电泳和流式细胞仪检测细胞凋亡。结果:TAM能抑制ER阳性和阴性乳腺癌细胞生长,阻滞细胞周期于G0／G1期,并可诱导细胞凋亡,相同浓度条件下,TAM 对ER阳性乳腺癌细胞的抑制作用强于ER阴性乳腺癌细胞:同时,TAM能拮抗外源性雌激素对MCF-7细胞的促生长作用,而对MDA-MB-231细胞的生长抑制作用与雌激素的存在与否无关; γ-干扰素预处理细胞24h后,TAM抗乳腺癌细胞的作用增强。结论:体外条件下,TAM有抗 ER阳性和阴性乳腺癌细胞作用,作用机制是通过影响细胞周期、诱导细胞凋亡,γ-干扰素能加强TAM的抗乳腺癌作用。更多还原

【Abstract】 Objective: To investigate the enhancing effect of γ-interferon on anticancer effect of tamoxifen against breast cancer cells in vitro. Methods: MCF-7 ER-positive and MDA-MB-231 ER-negative breast cancer cell lines were treated with tamoxifen alone, or in combination with γ-interferon and estrogen in vitro. Cell proliferation was evaluated by MTT assay, distribution of cell cycle were determined by flow cytometry, DNA gel electrophoresis and FCM were used to determine cell apoptosis. Results: The inhibitory effect of TAM on proliferation of breast cancer cells was associated with cell cycle arrest in G0/G1 phase and with induction of apoptosis, and at the same concentrations, the inhibitory effect of TAM on ER-positive cells was higher than that of ER-negative cells. Proliferation-promoting effect of estrogen on MCF-7 was blocked by TAM, but proliferation of MDA-MB-231 inhibited by TAM was not related to estrogen. Anticancer effect of TAM was enhanced when Cells were pretreated with γ-interferon for 24 hours. Conclusion: TAM has anti-cancer effect by inhibiting proliferation and inducing apoptosis in ER-positive/negative breast cancer cells in vitro, and γ-interferon can enhance anticancer effect of TAM on breast cancer cells.更多还原
