蒽酮-硫酸法测定出芽短梗霉发酵液残糖、总糖含量的研究

【摘要】 目的研究色素和不同比例乙醇对蒽酮-硫酸法测定出芽短梗霉发酵液残糖、总糖含量的影响。方法配制不同比例乙醇蔗糖溶液,蒽酮-硫酸法测定蔗糖含量;发酵液稀释300倍620 nm处测OD值;2倍乙醇沉淀发酵液,沉淀上清稀释100倍,620 nm处测OD值。结果纯化水:乙醇比例为1:1,1:2时可用蒽酮-硫酸法测其中蔗糖含量;发酵液稀释300倍,乙醇沉淀上清液稀释100倍后,620 nm处吸收值均接近0。结论蒽酮- 硫酸法可用于出芽短梗霉发酵液残糖、总糖测定。
【Abstract】 Objective To study the influence of pigmen and different ratio of ethanol on determination of the whole and residual sugars from the fermentation broth by the anthrone-sulfuric acid colorimetry method. Methods The content of sucrose solutions with different ratio of ethanol was determined by the anthrone-sulfuric acid colorim- etry method. OD value was determined at 620 nm from 300-fold dilution of the fermentation broth and 200-fold dilution of the supernatant which was obtained by ethanol deposit from the fermentation broth. Results The content of sucrose solutions with different ratio of ethanol (1:1,1:2) was detected by the anthrone-sulfuric acid colorimetry method. The value of OD620 from 300-fold dilution of the fermentation broth and 200-fold dilution of the supernatant were near to zero. Conclusion The anthrone-sulfuric acid colorimetry method can be used for the determination of the whole and residual sugars from the fermentation broth.
