重组瑞替普酶对体内诱发形成血栓的快速溶栓作用

【摘要】 目的通过建立以胶原蛋白-肾上腺素诱发小鼠体内血栓形成与角叉莱胶诱发大鼠尾部血栓形成模型,评价重组瑞替普酶(Reteplase)对体内诱发形成血栓的快速溶栓作用,为临床应用提供依据。方法将瑞替普酶与诱导剂混匀后给小鼠作尾静脉注射,观察给药后5min内小鼠死亡数和15min内小鼠偏瘫恢复数,计算药物对小鼠形成脑血栓的保护率;大鼠足跖皮下注射角叉莱胶使尾部形成血栓后给药, 观察血栓平均长度及再通率。结果和结论瑞替普酶对胶原蛋白-肾上腺素诱发小鼠体内血栓有迅速溶栓、急救效应;对角叉菜胶诱发大鼠尾部混合血栓有显著溶栓作用:溶栓作用呈现明显的剂量-效应关系。

【Abstract】 Objective To evaluate the rapid dissolution effects of Recombined Reteplate on blood clots in duced in vivo, and guide its clinical applications. Method Evaluate the rapid dissolution effects of Reteplate on blood clots. The blood clots models in mice were induced by collagen-adrenalin in vivo and the rat models were induced by carrageenin. Result and Conclusion Reteplase could rapidly dissolve the blood clots in mice induced by collagen-adrenalin and showed emergence treatment effects to mice. Also it was effective to the composed blood clots induced by carrageen in rat. The blood clots dissolution effects showed dosage-effect Relationship remarkably.
