LPS免疫激发大鼠视上核内一氧化氮表达的昼夜节律

【摘要】 目的探讨LPS免疫刺激时大鼠下丘脑视上核一氧化氮表达的昼夜节律变化。方法成年雄性SD大鼠24只随机分为4组,分别在16:00、22:00、4:00、和10:00时腹腔注射细菌脂多糖(LPS)。两小时后,分别灌注固定取脑。采用β-NADPH-酶组织化学和计算机图象分析技术检测视上核中NADPH-d阳性产物的光密度(OD)值,并利用余弦法分析这组数据的昼夜变化规律。结果四组大鼠下丘脑视上核内NADPH-d阳性产物的OD值显示明显的昼夜节律性,峰值在上午9:28。结论免疫激发状态下大鼠下丘脑视上核一氧化氮表达呈现昼夜节律性变化,SON可能通过NO途径参与免疫节律的调节。
【Abstract】 Objective To investigate the circadian rhythm changes of nitric oxide expression in supraoptic nucleus(SON) of immune challenge rats induced by LPS. Method A total of 24 adult male SD rats were randomly divided into 4 groups according to intraperitoneal injection with lipopolysaccharide (LPS) at16:00, 22:00, 4:00 and10:00 oclock respectively. Two hours later, all the rats were sacrificed after perfusion and fixation, then their brains were taken. NADPH-d histochemistry and image analysis method were used to detect the optical density (OD) value of NADPH-positive products in SON, and one-way ANOVO and Cosinor methods were applied to analyze the circadian rhythm of these data. Result The OD values of NADPH-d positive products in SON of the four groups of rats showed the obvious circadian rhythm with acrophase at 9:28 am. Conclusion NO expression in SON show significant circadian rhythm.in immune-irritated rats.
