褪黑素、胸腺肽、白细胞介素-2免疫调节作用的比较及褪黑素调节免疫功能的机制

【摘要】 目的 比较褪黑素(Melatonin,MT)、胸腺肽(Thymosin,THS)、白细胞介素-2(Interleukin 2,IL-2)的免疫调节作用及探讨褪黑素调节免疫功能的机制。方法 比较MT、THS及IL-2对环磷酰胺(Cyclophosphamide,CY)小鼠碳粒廓清能力、血清溶血素水平及迟发超敏反应的影响。运用纳洛酮(Naltrexone,NAL)、利血平(Reserpine,RESP)、酚妥拉明(Phentolamine,PHE)及普萘洛尔(Propranolol,PROP)等工具药,对MT调节免疫功能的机制进行了探讨。结果 对于CY小鼠非特异性免疫功能的增强作用,MT、THS及IL-2的效果相似;对于特异性免疫功能的增强作用,MT的效果较THS及IL-2全面。在体外,MT可显著增强ConA诱导的BABC小鼠脾淋巴细胞增殖反应;能明显增强CY小鼠IL-2的生成及活性;NAL、RESP、PHE及PROP能完全或部分阻断MT的免疫增强作用。结论 MT既可直接作用于免疫细胞,又可促进免疫细胞释放阿片肽、IL-2等细胞因子进而参与免疫调节作用。此外,MT还可以通过激活肾上腺素能神经系统α和β受体简介调节机体免疫功能。

【Abstract】 Objective To compare the effects of Melatonin(MT),Thymosin (THS) and Interleukin 2(IL-2) and investigate the mechanisms of immune-modulating function of MT. Methods The clearance rate of charcoal particles,production of serum hemolysin and delayed hypersensitivity reaction of MT,THS and IL-2 were compared in CY-treated mice. It was conducted to study the mechanisms of MT by using naltrexone,reserpine,phentolamine and propranolol. Results The effects of MT,THS and IL-2 on the indistinctive immune reaction in CY-treated mice were alike and the effects of MT on the distinctive immune reaction in CY-treated mice were complete. In vitro,MT remarkably increased ConA-induced lymphocyte proliferation of spleen cells from BABC mice. MT significantly enchanced the production and activity of IL-2 in CY-treated mice. The enhancing immune function of MT was completely or incompletely held back by NAL,RESP,PHE and PROP. Conclusion MT could not only act directly on the immune cells and stimulate the release of opioid peptides by activating the immune cells,but act indirectly on the adrenergic system to modulate the immune functions also.
