【摘要】 本文以史氏鲟鱼软骨为原料,按碱提—酶解—醇沉的工艺流程,进行提取硫酸软骨素的碱提工序工艺研究。通过单因素的不同碱浓度、碱液温度、碱液量以及该三个因素的L9(33)的正交试验,分别研究其对产品得率及其氨基己糖含量的影响,综合分析得出碱提工序的最佳工艺以2倍量6%的NaOH为溶剂,于40℃的水浴中搅拌提取4h;该工艺条件制备鲟鱼硫酸软骨素的得率可高达40.45%,质量符合WS1-C3-0030-2000(硫酸软骨素口服片)和WS-10001-(HD-0892)-2002(硫酸软骨素注射品)的标准,其红外光谱图也与标准品相似。鲟鱼硫酸软骨素精品和粗品对三种细胞乳腺癌细胞株(MCF-7细胞)、胃癌细胞株(MGc803细胞)、肝癌细胞株(SMMC7721)作用72h时其IC50分别为7.28mg/ml、7.68mg/ml、4.25mg/ml和8.86mg/ml、9.27mg/ml、7.37mg/ml,与鲨鱼硫酸软骨素精品和粗品有着相似的抗肿瘤活性。更多还原

【Abstract】 In this paper,the Amur sturgeon cartilaginous were taked as the material,the process flow alkali extraction—enzymatic hydrolysis—alcohol precipitation were used,the alkali extraction technology of chondroitin sulfate were studied.By single factors(different alkali concentration,alkali liquor temperature,alkali liquor content) and orthogonal test,the effect on yield of product and hexosamine content were studied,the optimum alkali extraction technology was obtained as follows:adding 2 times amount of 6%NaOH,stirring in water at 40℃for 4h,the yield of chondroitin Sulfate made by this technology reached to 40.45%,the quality met the requirement of WS1-C3-0030-2000 and WS-10001-(HD-0892)-2002,and its infrared spectrum were similar with standard sample.The breast cancer cell line(MCF-7 cell), gastric cancer cell line(MGc803 cell) and hepatoma cell line(SMMC7721) were treated with the pure and rough samples of sturgeon chondroitin sulfate 72h,and its IC50 were 7.28mg/ml、7.68mg/ml、4.25mg/ml和8.86mg/ml、9.27mg/ml、7.37mg/ml respectively,its antitumor activities were similar with the pure and rough samples of shark.更多还原
