花鳗鲡促性腺激素cDNA克隆与其在激素诱导性腺发育过程中的作用

【摘要】 鱼类促性腺激素(GtH)是一种由脑垂体分泌的糖蛋白激素,对鱼类性腺的发育成熟和生殖细胞的发生、分化、发育及排放都有重要的调节作用。鱼类脑垂体分泌产生两种促性腺激素(GtH-Ⅰ和GtH-Ⅱ),其中GtH-Ⅰ相当于四足类脊椎动物的促卵泡素(FSH);GtH-Ⅱ相当于四足类脊椎动物的促黄体素LH。GtH-Ⅰ和GtH-Ⅱ都是由α和β两个亚基组成,它们具有相同α-亚基和不同的β-亚基。我们利用GeneRacerTM技术,分别克隆了花鳗鲡两种促性腺激素的α和β亚基全长cDNA。其中,GtHα亚基cDNA全长为362bp,编码92个氨基酸;FSHβ亚基cDNA全长为462bp,编码105个氨基酸; LHβ亚基的全长cDNA为590bp,编码116个氨基酸;与其他鳗类的三种亚基氨基酸比对表明,花鳗鲡GtHα,FSHβ与LHβ亚基的半胱氨酸和潜在的N糖基化位点是极其保守的。运用realtime PCR技术检测了花鳗鲡脑垂体GtHα,FSHβ与LHβ在雄雌性腺发育过程中的表达变化。结果显示:垂体中GtHα,LHβ的表达均随着性腺发育逐渐升高,在雌鱼卵黄生成晚期,雄鱼排精期达到最高:垂体中FSHβ的表达在雌鱼卵黄生成前期,早期,中期均维持较高的水平,而在卵黄生成晚期和排卵前期有着显著的下降:在雄鱼精子发生早期FSHβ的表达量较高,在精子发生晚期与排精期则较低。上述结果提示FSH与LH在花鳗鲡的精子发生和卵子发生中起着重要的作用。
【Abstract】 The gonadotropin(GtH) belongs to the glycoprotein hormone family and play central roles in vertebrate reproduction.In teleost,like in other vertebrates,the gonads are stimulated by two gonadotropin secreted from the pituitary,follicle-stimulating hormone(FSH) and luteinizing hormone(LH),which are originally named as GTH-Ⅰand GTH-Ⅱ,respectively.They are heterodimeric,which consist of anα-subunit and the corresponding hormone-specificβ-subunit.In present study,three full-length cDNAs encoding the common glycoproteinαsubunit(CGα),FSHβand LHβsubunits,were isolated from marbled eel.The cDNA of CGαis 362-bp in length and encoded 92 amino acids(aa).The cDNA of FSHβconsisted of 462-bp, encoding a putative protein of 105 aa,while the cDNA of LHβconsisted of 590-bp,encoding 116 amino acids.Comparison of the deduced amino acid sequences of marbled eel CGa,FSHβ,and LHβwith that of other eels showed fully conserved of the cysteine residues and potential N-linked glycosylation sites were fully conserved among different eel species.Quantitative realtime RT-PCR analysis revealed that the CGαand LHβtranscriptional levels in the pituitary of female and male gradually increased during the artificially inducing gonadal development,and peaked at late vitellogenic stage and spermiation stage respectively. FSHβmRNA level in the pituitary of female was high at previtellogenic stage,early vitellogenic stage and mid-vitellogenic stage,but declined sharply at late vitellogenic stage and migratory nucleus stage.In male marbled eel,the mRNA level of FSHβin the pituitary was higher at early spermatogenesis stage than at late spermatogenesis stage and spermation stage.These results suggest that FSH and LH might be involved in regulation of spermatogenesis and oogenesis in tropic eel A.marmorata.
