迟钝爱德华氏菌(Edwardsiella tarda)粘附及侵袭特性研究

【摘要】 用分离自患病大菱鲆(Scophthalmus maximus)的迟钝爱德华氏菌(Edwardsiella tarda)L-49231菌株接种体外培养细胞(HaCaT细胞),观察其对细胞的粘附性和侵袭性,并研究了不同孵育时间对HaCaT细胞粘附性及侵袭性的影响,不同处理因子处理细菌对细胞的粘附性和侵袭性的影响。结果表明:L-49231菌株能粘附HaCaT细胞,对HaCaT细胞具低度侵袭力,并且细菌粘附细胞与侵入细胞的数量随作用时间的增加而增加。分别用甲醛、戊二醛、胰蛋白酶、盐酸处理L-49231菌体,细菌粘附力及侵袭力均有所下降;用蔗糖、甘露糖处理菌体,细菌的粘附力变化不大,用葡萄糖处理,能够促进细胞对细胞的粘附作用,侵袭实验结果表明,用廿露糖处理菌体,侵袭力变化不大,用蔗糖、葡萄糖处理菌体,能促进细菌对细胞的侵袭作用;用胰酶处理菌体,能抑制细菌对细胞的侵袭作用。用自制抗体处理L-49231菌体,细菌粘附力及侵袭力均有所下降。
【Abstract】 The adhesive speciality and invasion speciality of the L-49231 strain that was isolated from diseased Scophthalmus maximus and indentified to be Edwardsiella tarda were observed with being inoculated in 24-well cell culture plate with HaCaT cell.The influence of different action time and different factors to adhesive speciality and invasion speciality of the L-49231 strain were studied.The rusults indicate:L-49231 strain can adhere HaCaT cell and invade into it after 2 hours.The quantity of bacteria adhered and invaded to cell increased with the prolonged action time.The adhesive and invasion capability of L-49231 strain declined after treated with Formaldehyde,Glutaraldehyde,Hydrochloric acid,Trypsin; adhesive capability varied little in disposal of Sucrose and D-mannose,but increased with Glucose.The effect to invasion capability indicate:the rate of invasion to cell to rise when the strain L-49231 was dealed with Glucose and D-mannose;when dealed with Sucrose,the rate of invasion to cell was partially restrained,when dealed with antibody,both its adhesion and invasion ability declined distinctly.
