白斑综合症病毒(WSSV)对锯缘青蟹的致病性及免疫指标的影响

【摘要】 采用巢式PCR(nested PCR)对2006年收集的锯缘青蟹(Scylla serrata)病蟹样品,开展了白斑综合症病毒(white spot syndrom virus,WSSV)检测,阳性检出率为15%,且WSSV的检出高峰与锯缘青蟹发病高峰相吻合,提示WSSV与锯缘青蟹的疾病发生可能存在相关性。为进一步明确WSSV对青蟹的致病性,采用病毒匀浆除菌过滤液,以1: 20、1:200、1:2000和1:20000四个稀释度肌肉注射感染锯缘青蟹,表明WSSV对锯缘青蟹有较强的毒力,1:20、1:200组死亡率为100%;1:2000、1:20000组死亡率为67%。对感染死亡蟹进行了病毒检测、血淋巴主要酶指标测定,结果攻毒死亡蟹组织中可检测到WSSV;且其蛋白含量、超氧化物歧化酶(SOD)、酚氧化酶(PO)和过氧化物酶(POD)活力均比对照组明显降低,而谷丙转氨酶(GPT)和谷草转氨酶(GOT)的活力明显高于对照组,碱性磷酸酶(ALP)的活力比对照组略有升高。

【Abstract】 White spot syndrome virus(WSSV) is currently the most serious viral pathogen of aquacultured shrimp worldwide,which is also able to infect a wide range of aquatic crustaceans(including crabs).In this study,diseased mud crabs(Scylla serrata) collected in 2006 were detected for WSSV by the method of nested PCR with the positive rate of 15%. Healthy mud crab were challenged by intramuscular injection in WSSV filtrate at the 1:20, 1:200,1:2000 and 1:20000 dilution.The cumulative mortality for treatment of 1:20 and 1:200 both reached 100%at 3 and 5 days post-infection,respectively.For 1:2000 and 1: 20000 treatment,the mortality both reached 67%at 5 and 8 days post-infection,respectively. WSSV were detected in infected crabs while negtive in the control group.Some important hemolymph enzymes were measured in WSSV-infected and control group.The protein concentration,superoxidase dimutase(SOD),phenoloxidase(PO),peroxidase(POD) activities were markedly decreased in of WSSV-infected crabs,while the level of glutamic-pyruvic transaminase(GPT) and glutamic-oxaloacetic transaminase(GOT) were significantly higher than control group.In addtion,Palaemon carincauda collected from diseased mud crab ponds were also WSSV positive,implicating shrimp was probably the virus carrier.The results of study indicated WSSV was highly virulent and probably potential a causative pathogen to mud crab.
