细菌基因组DNA免疫对鳗鲡部分体液免疫因子的影响和抗感染作用

【摘要】 本文报道了利用嗜水气单胞菌(Aeromonas hydrophila)的基因组DNA作为免疫原,肌肉注射免疫欧洲鳗鲡(Anguilla anguilla),比较研究免疫后欧洲鳗鲡的部分体液免疫因子的变化及抗细菌感染能力。试验分空白对照组(A组)、生理盐水对照组(B组)、1μg/尾基因组DNA免疫试验组(C组)、2μg/尾基因组DNA免疫试验组(D组)和溶血素基因PCR产物免疫试验组(E组)和灭活菌苗免疫试验组(F组)等六组,比较免疫注射后第7天、14天和28天,各组之间血清溶菌酶活性、抗微生物活性、过氧化物酶活性、酚氧化酶活性、抗嗜水气单胞菌抗体和血清总蛋白含量的变化,并于免疫第28天进行人工感染试验,比较各组之间抗感染能力。研究结果:1)与空白对照组(A)和生理盐水对照组(B)相比,免疫后第28天,免疫试验组(C、D、E、F)血清溶菌酶和抗微生物活性均显著提高(P<0.05)。与空白对照相比,整个试验过程,免疫试验组(C、D、E、F)的血清过氧化物酶和酚氧化酶活性无显著性差异(P>0.05)。2)免疫后第28天,除E组外,免疫试验组(C、D、F)鳗鲡血清中抗嗜水气单胞菌抗体的检出率达100%。3)人工感染试验结果,除E组外,免疫试验组(C、D、F)的免疫保护率约为空白对照组2~3倍。结果表明,适当剂量的细菌基因组DNA免疫可提高鳗鲡抗病力。

【Abstract】 The genomic DNA extracted from Aeromonas hydrophila is used as the immunogen for intramuscular injection Anguilla anguilla.The control(A),physiological saline(B),1μg genomic DNA immunity(C),2μg genomic DNA immunity(D),the PCR product of haemolysin(E)and inactivation vaccine(F)were consisted in the experiment.Sera were collected at 7th,14thand 28thafter immunity injection.The changes of lysozyme activity,antimicrobial activity,peroxidase activity,phenol oxidase activity and the antibody to Aeromona hydrophila in the serum were analyzed.Anguilla anguilla were challenged with Aeromona hydrophilla after immunizing 28 days,then the survival rates were calculated. The result showed,at the 28thof post-injection,the genomic DNA immunity(C,D)and inactivation vaccine(F)to Anguilla anguilla,the lysozyme activity and the antimicrobial activity significantly enhance (P<0.05)in serum when compared with control(A)and physiology saline(B);antibody level significantly enhance(P<0.01)too,bechance rate of antibody achieve to 100%;the survival rates notably enhance about 2~3 times;the peroxidase and phenoloxidase activity not singificant difference in the test(P>0.05).
