青虾、海南沼虾及其杂种rDNA ITS区的序列分析

【摘要】 应用核rDNA的内转录间隔区(ITS)分析技术,对青虾、海南沼虾及其杂交后代进行DNA分子水平的遗传分析。分子量测定表明,青虾和海南沼虾扩增出来的ITS1片段,大约在1700bp左右,存在大量的重复序列,并且有微卫星DNA片段。青虾和海南沼虾碱基序列的种间差异十分显著,同时青虾存在着较大的种内变异。F1代个体既有来源于海南沼虾的序列,也有来源自青虾的序列,从分子水平证明了青虾和海南沼虾杂交的成功。海南沼虾、F1代个体和回交后代拥有一段完全相同的缺失,而与其它样品在该区段的缺失明显不同,说明F1代和青虾的回交也是成功的。结果表明,ITS分析技术在淡水沼虾中有很大的应用潜力。
【Abstract】 ITS sequences of nuclear rDNA of Macrobranchium nipponense,Macrobranchium hainanense, their hybrid and offspring of back cross were analyzed.There were about 1700bp from ITS 1,in which a large rannber of repeated sequences including some microsatellite DNA segments,were found.Sequences of ITS 1 showed sufficient differences between Macrobranchium nipponense and Macrobranchium hainanense,while,intraspocific divergences of ITS 1 sequences appered in Macrobranchium nipponense. In hybrid(F1)and progeny of back cross,both paternal and maternal sequences of ITS 1 were detected. Results of sequencing of ITS 1 demonstrated that the artificial hybridization between Macrobranchium nipponense and Macrobranchium hainanense,back crossing(their hybrid with their maternal parent)were successfully achieved.Became of its abundant variations,ITS sequence analysis will have a great potential application in genetic structure analysis of Macrobrachium.
