水体铜对中华绒螯蟹鳃和肝胰腺显微结构及金属硫蛋白含量的影响

【摘要】 采用生态学单因子梯度试验,观察在水体中添加0.00,0.01,0.10,L 00,5.00 mg/L的铜(Cu2+)对中华绒螯蟹鳃和肝胰腺组织显微结构的影响。结果表明,0.01 mg·L-1Cu2-处理组与对照组均无显著差异,随着水体中Cu2-浓度的增高,整个鳃叶出现不规则增厚,血腔增大,血细胞增多,上皮层破坏直至解体;肝胰腺肝小管单层柱状上皮细胞肿胀、解体,细胞核固缩、破碎,致使肝小管管腔变细;基膜呈波纹状拱起,甚至断裂,并与柱状上皮之间形成巨大的空泡;血窦腔中的血细胞增多,在基膜断裂处与游离的柱状上皮细胞核充挤在一起。采用镉饱和法研究水体铜对中华绒螯蟹肝胰腺金属硫蛋白含量的影响表明,0.01 mg/L Cu2-处理组与对照组无显著差异,随着水体中Cu2-的增高,金属硫蛋白在0.10 mg/L Cu2-时达到峰值。当Cu2-浓度继续增高时,金属硫蛋白含量无显著变化。本研究结果显示高浓度的Cu2-对中华绒螯蟹鳃和肝胰腺组织显微结构会产生明显的损伤,并可以诱导肝胰腺金属硫蛋白的含量增加。

【Abstract】 Effects of different water-borne copper(Cu2+)(0.00,0.01,0.10,1.00,5.00 mg/L)on the microstructure of gills and hepatopancreas in Eriocheir sinensis were studied by Single Gradient Factor Experiments.The results showed between the 0.01 mg/L Cu2+group treated and the control group,the microstructure is of no significant difference.With the increasing of Cu2+level in the gill it was found irregularly thickened lamella and enlarged haemocoels increased hemocytes and damaged or disassembled epithelia.In the hepatopancreas because the simple columnar epithelia were intumesced and even destroyed,the spaces of the small blind tubules decreased And nuclears of the epithetlia were karyopycnosis or karyorhexis.The basement membranes were rumpled or even ruptured,and vacuoles were observed between the basement membrane and the simple columnar epithelia.The increased hemocytes in haemocoels was noted and they piled together with the dissociative epithetlium where the basement membranes were ruptured.The effect of Cu2+on the content of metallothionein was also investigated by Cadmium-Saturation Methods.There was no significant difference between the control group and 0.01 mg/L water-borne Cu2+group,.The maximum content of metallothionein was achieved when the water-borne Cu2+was 0.10 mg/L.This content was almost kept even Cu2+level was further increasd.Results of the present study indicated that high level water-borne Cu2+impaired microstructure of the gills and hepatopancreas obviously,and induced the increase of metallothionein in the hepatopancreas.
