氨甲蝶呤明胶微球的研制及相关生物学特性

【摘要】 目的研制包裹有氨甲蝶呤的颗粒型可降解性栓塞剂。方法采用改良的双相乳化冷凝聚合法制备氨甲蝶呤明胶微球,并测试微球的包裹率、载药率及体外释药特性。结果氨甲蝶呤明胶微球颗粒直径均匀,药物包裹率85%,载药率32.2%,体外4h内缓释85%。结论本法制得的氨甲蝶呤明胶微球性质稳定,微球降解速率及药物释放速度基本符合临床要求。

【Abstract】 Objective To study a kind of biodegradable embolic agent encapsulated with amethopterin.Methods Amethopterin Gelatin microspheres were prepared by improved emulsified cold-condensation method, the encapsulation rate and drug content tested, also the characteristics of drug release evaluated.Results The diameter of amethopterin Gelatin microspheres could be effectively controlled. The encapsulation rate was 85%, and drug content was 32. 2%. 85% of the drug was sustained-release in 4 hours.Conclusion Amethopterin Gelatin microspheres prepared by this method were stable; the velocity of gelatin decomposition and drug release was applicable for clinical use.
