甲基莲心碱对动脉粥样硬化相关病变的影响及其机制研究

【摘要】 目的本实验旨在研究甲基莲心碱(neferine,Nef)对天然及氧化修饰的低密度脂蛋白导致巨噬细胞脂质堆积、增殖的影响及其作用机制的探讨。方法以琼脂糖电泳、TBARS值为指标,鉴定Cu2+介导的LDL氧化。以LDL及Ox-LDL为泡沫细胞诱导剂,观察Nef对巨噬细胞系RAW264.7细胞内TG、TC堆积的影响,通过测定细胞内超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)的活性探讨其机制。观察Nef对Ox-LDL导致的RAW264.7细胞增殖的影响,通过测定胞内[Ca2+]i探讨其可能机制。结果Nef可抑制LDL、Ox-LDL引起的RAW264.7细胞内TG、TC的堆积,可抑制Ox-LDL诱导的RAW264.7细胞的增殖,可提高RAW264.7细胞内SOD和GSH-PX的活性,可降低RAW264.7细胞内[Ca2+]i。结论Nef可能通过增强细胞自身抗氧化能力和钙离子拮抗作用影响动脉粥样硬化(Atherosclerosis,AS)发生发展的多个环节,对防治AS具有一定的作用,有广泛的开发前途。

【Abstract】 Objective We investigated the effects and mechanisms of neferine ( Nef) in anti - foam formation and anti - proliferation of macrophages. Method The thiobarbituric acid - reactive substance (TBARS) values and relative electrophoretic mobility were determined to evaluate oxidatively modified degree of LDL induced by copper. RAW264. 7 cells were exposed to different concentration of LDL and ox - LDL, and incubated with different concentration of Nef, respectively. The tryglyceride (TG) and total cholesterol (TC) content in the cells were assayed. SOD activity, GSH - PX activity and the content of intercellular Ca2 + in RAW264. 7 cells were detected. Result Nef can inhibit accumulation of lipids stimulated by LDL and ox -LDL. LDL and ox - LDL can decrease the SOD activity and GSH - PX activity in the macrophages and Nef can reverse the SOD activity and GSH - PX activity induced by LDL and ox - LDL. The similar results were gained for the content of Ca2 + in the macrophages. The results also showed that Nef inhibited macrophages proliferation induced by ox - LDL. Conclusion These data suggest that Nef can influence multiple functions in the process of anti - atherosclerosis by anti - oxidation and anti - proliferation.
