农杆菌介导法获得彩色棉转基因植株的研究

【摘要】 采用6个不同基因型彩色棉作为外植体,通过农杆菌介导法将伸展蛋白基因(Expansin)导入彩色棉,成功获得转基因植株。研究了不同基因型对愈伤组织诱导率和分化率的影响。筛选出易分化受体基因型绿彩棉。试验还筛选出再生植株的生根选择压。将获得的38个株系的转基因植株在卡那霉素筛选培养基上进行生根筛选。结果显示,有13个转基因株系10天左右在含有卡那霉素100mg·L-1培养基上生根,形成转化植株,而非转化植株完全不能生根。DNA Southern blot分子检测结果证实,绿彩棉再生植株实现了外源基因的成功转化,为彩色棉遗传转化的进一步研究工作奠定了基础。
【Abstract】 In this study, cell extensin genes were introduced into 6 different genotypes of colored cotton as explant receptors by Agrobactirum-mediated transformation and transgenic plants have been successfully obtained. We studied the effect of different genotypes on callus inductivity and differentiation efficiency. A highly differential acceptor genotype of green-colored cotton was gotten. The rooting selection pressure of regenerated plants was determined in the experiment, transgenic plants of 38 lines were selected with kanamycin 100 mg·L-1 pressure by roots information. The result showed that transgenic plants from rooted 13 lines on the selection pressure’s medium about ten days. Southern-blot analysis confirmed that foreign gene had been already integrated into the cotton genome. The transgenic plants obtained and the genetic transformation system established in this experiment were available for further colored-cotton genetic transformation
