光皮桦天然群体遗传多样性分析

【摘要】 利用RAPD标记技术对福建省9个光皮桦天然群体135个个体的遗传多样性和群体遗传结构进行了分析。结果表明:21个随机寡核苷酸引物共检测到225个位点,其中多态位点203个,占90.22%;物种水平的Shannon多样性指数I=0.4908,Nei’s基因多样度h=0.3323;综合评价9个样本群体,以永安(YA)群体的DNA水平遗传多样性最高,其次是武平(WP)、沙县(SX)、连城(LC)、武夷山(WY)、明溪(MX)、邵武(SW)、尤溪(YX),而沙县罗卜岩(SL)群体拥有最低DNA水平的遗传多样性。分子方差分析(AMOVA)结果表明,光皮桦群体的遗传变异主要存在于群体内(占71.31%),群体间变异组分仅占28.69%,这与利用Shannon多样性指数估算的分化系数(I_T-I_S)/I_T=0.3250和Nei’s遗传分化系数Gst=0.2809的结果相似。通过UPGMA聚类分析将9个群体分为3个不同类群。

【Abstract】 Betula luminifera is a fast growing deciduous broad-leaved tree species endemic to China.The remnant populations are generally fragmented and highly disturbed because of exploitation for timber and other species invasion.In this study,the genetic diversity and population genetic structure in 135 trees from 9 natural populations of B.luminifera were assessed using RAPD markers.21 RAPD primers amplified a total of 225 scorable markers,203 of which(90.22%)were polymorphic at species level.The genetic diversity,estimated by Shannon’s index and Nei’s gene were 0.4908 and 0.3323 at the species level, respectively.There were the highest level of DNA genetic diversity in YA population and the lowest level of genetic diversity in SL population among 9 populations based on integration estimated.Analysis of molecular variance(AMOVA)indicated that 71.31%of the total genetic diversity distributed within population as well as 28.69%was observed among populations,in accordance with the result of POPGENE 1.32 revealing that genetic differentiation(I_T-I_S)/I_T and coefficient of gene differentiation(Gst)were 0.3250 and 0.2809,respectively.The UPGMA cluster analysis showed that three groups could be identified according to the nine populations of B.luminifera.
