尾叶桉无性系3228遗传转化体系建立的初步研究

【摘要】 采用根癌农杆菌介导法进行nptⅡ基因对广西桉树优良主栽品种尾叶桉无性系3228(Clone 3228 of Eucalyptus urophylla)茎段的遗传转化研究,对转化过程中的一些因素进行了探讨:桉树茎段对卡那霉素的敏感实验,得出最适筛选浓度为75mg/L,头孢噻肟钠的最适抑菌浓度为300mg/L;2-4min是最适合叶片侵染的时间;共培养2d对茎段转化较适宜,得到抗卡那霉素的抗性芽的频率为2.6%。
【Abstract】 stem of Clone 3228 of Eucalyptus urophylla containing nptⅡwas investigated and Agrobacterium tumifaciens-mediated transformation was used.Factor on Agrobacterium tumifaciens-mediated gene transfer was discussed indetail.The optimal concentration of kanamycin to select was 75mg/L and of cefotaxime sodium to strain fungus was 300mg/L.The pre-cultivation was unnecessary.The ideal time for stem dipping in Agrobacterium was 2-4 min and the optimal co-cultivation was 2 days.The ratio of Kanamycin-resistant shoots was 2.6%.
