闽楠群体遗传多样性RAPD分析

【摘要】 利用RAPD分子标记对闽楠分布于江西和福建的8个天然群体共计240个个体的遗传多样性和群体遗传结构进行了分析。15个多态引物共检测到143个位点,其中多态位点136个,占95.10%。物种水平Shannon多样性指数I=0.5028,Nei基因多样度h=0.3360,种内总遗传变异量Ht=0.3360,群体内遗传变异量Hs=0.1332,群体间变异量大于群体内变异量,表明闽楠的遗传变异主要存在于群体间。闽楠的遗传多样性较高,遗传分化系数Gst=0.6036,说明闽楠群体间的遗传变异占总变异的60.36%,群体间已出现强烈的遗传分化。生境破碎、群体隔离等可能是造成群体间遗传分化严重的原因。根据闽楠的遗传变异特点,初步提出了闽楠的保护策略。

【Abstract】 The genetic diversity and population genetic structure of 8 populations with total of 240 individuals in P.bournei distributed in Jiangxi and Fujian province were estimated by RAPD markers.The results demonstrated that 143 loci were amplified with 15 oligon-nucleotide primers, of which 136(95.1%)loci were polymorphic.Shannon’s information index(I)and Nei’s gene diversity(h)were 0.5028 and 0.3360 respectively at the species level.Total genetic diversity(Ht) was 0.3360,and the genetic diversity within populations(Hs)was 0.1332.The coefficient of genetic differentiation(Gst)were 0.6036,it means that the genetic variation among populations accounted for 60.36%of the total.This indicated that the genetic variation existed mainly among populations rather than within populations in P.bournei,and the gene flow(0.3284)among populations was very restricted.The reasons may be attributed to the changes of environment, habitat destruction and excessively making use of the species resources by local people.Based on above results,the conservation strategies of genetic diversity for the species were provided.
