凝胶型W/O/W多重乳液担载胰岛素的前期研究

【摘要】 胰岛素是治疗糖尿病的主要药物,目前临床上主要为皮下注射胰岛素给药,给患者带来极大的痛苦并伴有多种不良反应。胰岛素口服给药可以解决这些不便。本文采用多重乳液载药体系作为胰岛素的载体,创新性地将外水相凝胶固化来克服多重乳液的不稳定性问题,通过正交实验及单因素分析,重点考察了石蜡-span-tween乳液体系,确定了制备乳液过程中影响该乳液体系粒径大小及粒径分布的主要因素,得到了随各因素水平变化乳液液滴平均粒径、粒径分布的变化趋势。实验表明,当亲油相表面活性剂选用14 wt%span80与3 wt%Abil EM90复配、亲水相表面活性剂选用4%tween80、第一步制乳温度为50℃、内水相与油相体积比为4:5、原始乳液与外水相体积比为1:2时,所制得的乳液平均粒径为14.86 μm,粒径分布均匀,稳定性可达30天。在外水相中加入高分子增稠剂后,乳液的稳定性可进一步增强。

【Abstract】 Insulin is one of the most important drugs to cure diabetes. Presently, the insulin is mostly given to patients by injection in clinic, and too much injection per day causes patients pain and some accompanying side effects. The problem can be solved by oral administration of insulin. In this paper, W/O/W multiple emulsion is investigated to loaded insulin and an original idea gelled multiple emulsion is put forward. Through orthogonal experiment design and single parameter analysis, the main factors that influent the droplet size and distribution of the Liquid Paraffin-span80-tween80 are determined in the process of multiple emulsion preparation, and the variation of mean size, size distribution and incremental ratio changing with different factors and ranges is also determined. The experimental results indicate that homogeneous distribution and good stable W/O/W multiple emulsion with the mean size 14.86μm can be obtained when the lipophilic surfactant is the mixture of 14wt% span80 and 3wt% Abil EM90, the first stage of emulsification is carried out at 50℃, the primary emulsion consisted of 44.4% aqueous and 55.6% oily phase, the W/O/W multiple emulsion consisted of 33.3% primary emulsion and 66.7% of an aqueous solution containing 4wt% tween80. The stability will be enhanced when macromolecule gel is added.
