变形链球菌葡糖基转移酶植物表达质粒p2355-gtfB、p2365-gtfB转化烟草的研究

【摘要】 目的:将携带变形链球菌葡糖基转移酶编码基因gtf8的植物高效表达载体p2355-gtfB、p2365-gtfB转化到烟草植株中, 为研制有效的转基因植物防龋疫苗打下基础。方法:利用根癌农杆菌EHA105介导转化烟草,经抗性筛选获得抗性烟草植株后, 进一步对抗性植株进行CUS染色和PCR扩增等分子检测。结果:抗性筛选得到抗性烟草植株;GUS染色以及目的基因片段的 PCR扩增证实获得获得携带目的基因的阳性植株;结论:植物高效表达载体p2355-gtfB、p2365-gtfB已经成功转化到烟草植株中。
【Abstract】 OBJECTIVE: To transfer the high effective plant expression plasmids p2355-gtfB. p2365-gtfB earning gtfB gene of Streptococccus mutatis into the tobacco, and provide basis for further study on edible plants vaccine against dental caries. METHOD: Tobacco plant cotyledon were transformed by co-cultivating leaf disks method with Agrobacterium strains carring plant expression plasmids p2355-gtfB or p2365-gtfB. RESULTS: After selection with antibiotic. GUS assay and PCR amplification, the foreign gene gtfB was successfully transformed in tobacco. CONCLUSION: The transgenic tobacco were acquired and the transferred seedling can be selected using quick method.
