【摘要】 <正>目的:检测SD大鼠口腔颊粘膜鳞癌细胞系SDSCC-B与其单克隆细胞系Rca-B细胞的端粒酶的活性, 分析其对细胞转移能力的影响。方法:采用单细胞克隆培养法筛选细胞系SDSCC-B建立单克隆来源的细胞系Rca-B;分别检测两细胞系端粒酶的活性;以Tca8113和Tb细胞系做阳性对照。两细胞系裸鼠尾静脉注射后观察其实验性肺转移情况。检测结果采用SAS6．12统计软件包进行统计学分析。结果:端粒酶活性检测结果显示两个细胞系的端粒酶活性均为强阳性,显著高于大鼠正常颊粘膜细胞,差别均具有更多还原

【Abstract】 PURPOSE: To study the relationship of the telomerase activity and the potential of experimental metastasis to the lung between Sprague-Dawley rat buccal squamous cell carcinoma cell line and it’s purified sub-cell line. METHODS: In vitro inbred line Rca-B was been purified from SDSCC-B cell line by using mono-cell cloning method. The telomerase activity of cell lines was analyzed by TRAP-ELISA. The ratios of LW/BW (lung weight/body weight)of nude mice after experimental lung metastasis were to observe the metastatic characters. The results were analyzed by SPSS 11.0.1 statistical software. RESULTS: The findings showed that telomerase activity of the cell lines was higher than normal buccal mucosa cells with significant difference ( P<0.01 ) . telomerase activity of Rca-B cells was higher than that of SDSCC-B cells ( P<0.05 ) . The ratios of LW/BW of nude mice of the two cell lines were significantly different ( P<0.05 ) . CONCLUSION: Rca-B and SDSCC-B cells were high metastatic character, and Rca-B was more easily metastatic and lower differenciation .The expression of telomerase activity was relevant with metastatic ability of these cells.更多还原
