【摘要】 <正>目的:探讨基质金属蛋白酶-2活性与成釉细胞瘤体内外侵袭的关系及其机理。方法: 通过MMP-2 抑制剂Ro31-9790抑制MMP-2活性进行干预处理,采用成釉细胞瘤细胞原代培养、瘤组织块裸鼠肾包膜下移植、MTT、Western blotting、体外侵袭试验、组织学和免疫组织化学等方法,观察MMP-2活性与成釉细胞瘤体内外侵袭的关系并研究其机理。结果: Ro31-9790各处理组和对照组的生长曲线无显著差异

【Abstract】 Objective: To investigate the relationship between matrix metalloproteinase-2 ( MMP-2 ) activity and invasion of ameloblastoma and the mechanisms in vivo and vitro. Methods: The cells and xenograft of ameloblastoma were treated with matrix metalloproteinase-2 ( MMP-2 ) inhibitor Ro31-9790, and the effects of Ro31-9790 on the invasion of ameloblastoma were observed. Primary culture in vitro, subcapsular kidney xenograft in vivo, MTT assay, Western blotting, in vitro invasion assay, histochemistry and immunohistochemistry were employed to study the effects of invasion produced by Ro31-9790. Results: Ro31-9790 didn’t influence the growth of ameloblastoma cells, its cell number had no significant change, P > 0.05. Ro31-9790 could inhibit the invasion of ameloblastoma in vivo and vitro, and could increase the positive expression rate of collagen IV and decrease the abnormal expression rate of epithelial cadherin in xenograft of ameloblastoma, but could not influence the expression of MMP-2 in vivo and vitro. Conclusion: MMP-2 activity could directly contribute to the invasive capacity of ameloblastoma. Ro31-9790 can inhibit the invasion of ameloblastoma. Activated MMP-2 might influence the invasive capacity of ameloblastoma by regulating expression of collagen IV and epithelial cadherin.
