【Abstract】 Objective: To investigate the relationship between matrix metalloproteinase-2 ( MMP-2 ) activity and invasion of ameloblastoma and the mechanisms in vivo and vitro. Methods: The cells and xenograft of ameloblastoma were treated with matrix metalloproteinase-2 ( MMP-2 ) inhibitor Ro31-9790, and the effects of Ro31-9790 on the invasion of ameloblastoma were observed. Primary culture in vitro, subcapsular kidney xenograft in vivo, MTT assay, Western blotting, in vitro invasion assay, histochemistry and immunohistochemistry were employed to study the effects of invasion produced by Ro31-9790. Results: Ro31-9790 didn’t influence the growth of ameloblastoma cells, its cell number had no significant change, P > 0.05. Ro31-9790 could inhibit the invasion of ameloblastoma in vivo and vitro, and could increase the positive expression rate of collagen IV and decrease the abnormal expression rate of epithelial cadherin in xenograft of ameloblastoma, but could not influence the expression of MMP-2 in vivo and vitro. Conclusion: MMP-2 activity could directly contribute to the invasive capacity of ameloblastoma. Ro31-9790 can inhibit the invasion of ameloblastoma. Activated MMP-2 might influence the invasive capacity of ameloblastoma by regulating expression of collagen IV and epithelial cadherin.更多还原
