组织工程化人工涎腺样组织体外构建的初步研究

【摘要】 目的通过体外颌下腺细胞/材料复合体的培养,探讨在动物体内构建涎腺样人工组织的可能性。方法:将体外培养5天的颌下腺细胞/支架材料复合物植入SD大鼠面颊部,分别于1、2、4周取出复合体进行组织学及免疫组织化学的观察。本实验所有支架材料为自制的脱细胞SD大鼠和家兔气管。结果:1、2、4周后SD体内复合体上的细胞为涎腺上皮细胞。结论:附着在支架材料上植入SD大鼠体内的细胞分化良好。

【Abstract】 Objective The purpose of this study was to obtain submandibular gland cells (SSG) /scaffolds in vitro, and to investigate the possibility to reconstruct a tissue engineering artificial salivary gland organoid. Methods The 5-day cultured SSG/scaffold complexes were implanted adjacent to cheek skin of SD rats. The complexes were harvested after 1, 2, 4 weeks. The morphological and immunohistochemical observation were studied. The scaffolds used in this experiment are acellular matrix from tracheae of rabbits and SD rats Results The cells attached on scaffolds can be proved as salivary epithelial cells by morphological and immunohistochemical detection after 1,2,4 weeks. Conclusion The cells attached on the scaffolds that been implanted in the SD rats had ability of differentiation.
