多聚赖氨酸对胎鼠颌下腺细胞培养的影响

【摘要】 目的:研究多聚赖氨酸做为生长底物对体外培养胎鼠颌下腺上皮细胞生长的影响。方法:以多聚赖氨酸包被培养板,对颌下腺上皮细胞进行分离培养,应用相差显微镜及电镜观察对细胞生长特性,同时用免疫组织化学方法对细胞来源进行鉴定。结果:颌下腺细胞在包被有多聚赖氨酸的培养皿表面生长分化良好,呈单层生长;免疫组织化学检测Anti-Pan cytokeratin、Anti-EMA呈阳性表达。结论:多聚赖氨酸在体外培养的过程中,可以促进细胞的黏附与生长。

【Abstract】 Objective: To study the effect of polylysine(PL) coated on the cell survival and outgrowth of submandibular gland cells from embryonic rat cultured in vitro. Methods: The submandibular gland was isolated from embryonic rats and digested into dissociated cell suspention for culture with PL coated as substrate. The cells were observed with phase contrast microscopy and electron microscopy. Immunohistochemistry staining was used to determine the cells. Results: The cells cultured are polygon epithelium cells with high survival rate and they were positively stained with Anti-Pan cytokeratin and Anti-EMA, which were epithelial specific antibody. Conclusion: PL could promote the cells cultured in vitro attachment and living.
