灵芝孢子粉对NOD小鼠自发性涎腺炎的作用及机制

【摘要】 <正>目的:观察全破壁灵芝孢子粉(Ganoderma Lucidum spores,GLS)预处理对非肥胖性糖尿病 (nonobese diabetic mice)NOD小鼠自发性涎腺炎发病的影响及其在免疫系统和细胞凋亡方面的作用机制。材料与方法:选取5周龄雌性NOD/Lt小鼠及相应对照BALB/c小鼠共64只,分为8组, 每组8只。分别经胃灌注1g·kg-1·d-1、0.1g·kg-1·d-1灵芝孢子粉,连续10周,阳性对照为地塞米松 (DEX)、阴性对照为生理盐水(NS)。在15周龄检测刺激全唾液流率(total saliva flow rate,STFR), 处死小鼠取下颌下腺(SMG),电镜观察超微结构;HE染色比较各组自发性涎腺炎发病率,图像分
【Abstract】 Objectives: To observe the prevention effects and mechanism of Ganoderma Lucidum spores (GLS) on sialoadenitis in non-obese diabetic (NOD) mice. Materials and Methods: 64 of 5-week NOD/Lt mice and BALB/c mice were divided into 8 groups: 1g· kg-1 ·d-1、 0.1g·kg-1 ·d-1 of GLS group, dexamethasone (DEX) group and normal saline (NS) group, they were treated by gastrogavage daily for 10 weeks. Stimulated total flow rates (STFR) were tested, the incidence of sialoadenitis was tested by HE staining, quantification of serum IgG antibody was done by ELISA, and peripheral subsets of and apoptosis of T、 B lymphocytes were observed by flow cytometry. Apoptosis cells in submandibular gland (SMG) were tested by TUNEL method. Results: It showed that 1g· kg-1 ·d-1 of GLS decreased the degree of the lymphocytic infiltration of SMG, increased STFR, decreased the level of serum IgG through increased the apoptosis of B lymphocytes, and decreased the ratio of CD4+ and CD8+ T lymphocyte subsets than those of the NS group through decreased apoptosis of them at different levels. High-dose of GLS also can decrease cell apoptosis and increase the secretary granules in SMG Conclusions: 1g· kg-1 ·d-1 of GLS might relieve the sialoadenitis of NOD mice, the effects were related to its action on immune-regulating of the serum IgG level and the ratio of T, B lymphocytes subsets. It proves preventive effects of GLS on the onset of sialoadenitis of NOD mice.
