光动力疗法选择性损伤鸡冠血管特异光源研制及实验研究

【摘要】 目的:研制一种特异性治疗光源,用于选择性光动力损伤鸡冠血管,观察其对血管的闭锁能力。方法:研制出发射光主峰与光敏剂叶绿素衍生物4号吸收峰一致的特异治疗光源并检测其性能。该光源用于光动力损伤鸡冠血管,通过大体及组织学观察,评价血管损伤情况。

【Abstract】 PURPOSE: To develop a specific optic source for the selective photodynamic damage of capillaries in chicken comb and evaluate its vasoocclusive potential. METHODS: To develop an optic source of which the main emitting peak corresponds with the absorbing peak of Chlorophyll derivative (CPD4) and determine its characterization. Then the specific optic source was used for the photodynamic damage of blood vessels in chicken comb. The specimens were harvested and evaluated by gross and histological observation. RESULTS: The specifications of the optic source were as follow: wavelength 600 to 700nm with the main emitting peak at 650nm; power density: 300mw/cm2,250 mw/cm2,200 mw/cm2,150 mw/cm2,100 mw/cm2,50 mw/cm2; field size: 50mm.Gross observations showed blanching of the chicken comb. Histological changes included: erythrocytes and endothelial cell (EC) injury, vascular occlusion and rupture, hemorrhage and denaturation of perivascular collagen fiber. The normal overlying epidermis was completely intact. The mean penetration of the CPD4 sensitized PDT was approximately 2.0mm.CONCLUSIONS: The developed optic source had been successfully used for the photodynamic vascular occlusion of chicken comb without causing damage to adjacent structures. It may offer a promising approach to the treatment of vascular lesions in the skin of humans.
