【摘要】 <正>目的:建立一个稳定的人正常口腔粘膜角化细胞体外培养体系。方法:取正常口腔粘膜经 Dispase及胰蛋白酶消化获取粘膜角化细胞,采用角化细胞培养液进行原代及传代培养,并进行形态学观察和角蛋白免疫组化染色等一系列细胞定性研究。结果:获取的细胞为纯的粘膜角化细胞; 细胞可传4-5代,成活40-50天;细胞呈铺路石状,电镜下角化细胞具有丰富的张力丝和桥粒等特更多还原

【Abstract】 Objective: To establish a method of culturing normal human oral keratinocoytes. Methods: Specimens obtained from healthy humans undergoing oral surgery were isolated by dispase and trypsin. Cells were grown in keratinocytes growth medium. The morphological characteristics were studied by light microscope and electron microscope. By immunohistochemistry, it proved there was cytokeratin in the cells. Results: The isolated cells were pure keratinocytes, which were subcultured for forth of fifth. There were demosomes between polygonal cells. The cells were positive to antibodies to cytokeratins. Conclusion: The culture technique for growing normal human oral keratinocytes was established.更多还原
