【Abstract】 Adenoid cystic carcinoma (ACC) of human salivary gland is well known as the characters of higher local invasion and relapse. Therefore, prognostic factors are urgently necessary to be learned for the clinical work. Object: The present study was to investigate the expression of the CD44s, CD44v6 and E-cadherin, α-catenin, β-catenin in odenoid cystic carcinoma (ACC) and ACC2,2 ACC-M cell line of the salivary glands and to reveal the relationship between the expression of the proteins and the tumor biological characteristics as well as the prognosis of ACC. Methods: The expression of adhesion molecules, which included, CD44s, CD44v6 and E-cadherin α-catenin, β-catenin, on 10 cases of normal salivary glands, 50 cases of human salivary ACC with various subtypes and biological features as well as two ACC cell lines and SCID mice neoplasm models were investigated by immunohistochemisty, and morphological. Results: In the clinical studies, CD44s, CD44v6 and E-cad-cat complex expression showed strong and stable expression in normal salivary glands tissues without exception. AT the same time the ruduced or complete loss of adhesion molecules expression, including CD44s, CD44v6 and E-cad-cat complex, on ACC cells has been observed. There was a significant difference in the proportion of cells staining posivtively for CD44s between the recurrent and the primary specimens (P<0.01). Down-regulation of E-cad/cat complex had been detected in 21 (42%) ACC cases, which associated with a more aggressive phenotype on nerve. Ultrastructure studies in SCID mice showed that there were more cytoskeleton components and cell superficial structures such as pseudo-process, microvillus, tight conjunctions and complexes of conjunctions in ACC-M cell. The expression of adhesion molecules in lung metastatic foci of ACC-M was higher than them in its subcutaneous tumor mass. We observed that the growth of ACC-M lung metastatic foci cells were slower than the cells of subcutaneous tumor and the mass in abdominal cavity, and the differentiation of former were higher than the latter. Conclusion: The clinical results revealed that: 1.Adhesion molecules HkeCD44s and E-cad are sort of important transmembrane proteins, which can maintain the normal structures in the salivary gland tissues. 2. Down-regulation of creation adhesion molecules shows to correlate with a higher propensity of tumor cells to detach from the primary site and to spread. 3. The loss of CD44s expression is contributed to the recurrence of human salivary ACC. 4. Down-regulation of E-cad/cat complex is associated with a more aggressive phenotype on nerve and vasculature. The animal studies revealed that: 1. The characters of ACC-M cells including cell differentiation, cytoskeleton and superficial linkage structures are facilitated for tumor metastasis and invasion. 2.The expression of CD44s in ACC cells in influenced by cell differentiation, and may effect the growth conditions. 3.
