大鼠骨髓基质细胞移植修复脊髓横断损伤的实验研究

【摘要】 目的:研究骨髓基质细胞修复脊髓损伤的可能性,探索治疗脊髓损伤的新方法。方法:取20只大鼠,制作椎体水平脊髓离断伤的大鼠动物模型,随机分为细胞移植组和单纯损伤组。前者将体外培养至第5代的大鼠骨髓基质细胞,用 Brdu 标记后,移植到大鼠脊髓离断处, 后者仅注射等量生理盐水。饲养12w,定期观察动物体重变化,进行功能评定,12w 后处死动物, 行病理,免疫组化和电镜检查。对比两组是否差异。结果:两组动物各项指标均有显著性差异。骨髓基质细胞移植组大鼠脊髓功能恢复优于单纯损伤组,大体所见及病理,免疫组化,电镜检查证明该组离断脊髓有明显修复迹象。结论:体外培养的大鼠骨髓基质细胞移植在大鼠骨髓内能够至少存活12w,并在脊髓损伤处发挥一定的修复功能。
【Abstract】 Objective:To study the extracting and culturing in vitro condition of the bone marrow stromal cells of the SD rats and observe the growing propert for their application on cell transplatation.Method:The bone marrow cells of the SD rats were separated and cultured.The cells sticking to the tube were transfered and purified to the bone marrow stromal cells.We observed their growing propert,drew their growth curve and estimate their split index and sticking rate.Result:The growing character of the cultured cells were stable before the 15thgeneration and the growth curves were similar.The split index of the 4thday was highest(18%).The sticking rate was the highest(90%) 14h after transfer.Conclusion:Acording to our study,the cultured bone marrow stomal cells were stable in vitro in several generations.They proliferated rapidly and had strong adaptation.The cultured bone marrow stromal cells could be the experimental cells for the cell transplantation.
