采用组织芯片技术检测乳腺癌中Skp2和p27的表达
【摘要】 目的观察乳腺癌中S期激酶相关蛋白2(S-phase kinase associated protein 2, Skp2)、细胞周期调控因子p27的表达情况,探讨它们在乳腺癌中表达的意义及相互关系。方法选取外科手术切除乳腺癌组织标本80例,采用组织芯片技术和免疫组织化学方法检测乳腺癌申Skp2和p27的表达。结果 80例乳腺癌中Skp2和p27的阳性率分别为46.3%、41.3%。 p27阳性乳腺癌中Skp2阳性率为15.2%,p27阴性乳腺癌中Skp2阳性率为68.1%,二者具有显著性差异,P<0.01;结论 Skp2、p27和Ki-67的表达均与乳腺癌组织学类型无关。Skp2 高表达、p27低表达与乳腺癌的浸润进展程度密切相关。Skp2高表达、p27低表达与乳腺癌淋巴结转移密切相关。乳腺癌中Skp2的高表达与p27的低表达之间明显负相关。在乳腺癌细胞增殖调控机制中,p27具有抑制细胞增殖的作用,Skp2促进细胞周期调控因子p27的降解,从而促进细胞增殖。应用组织芯片大规模高效检测临床组织样本是可行的,具有快速、方便、经济、准确的特点。
【Abstract】 Objective To study the expression of Skp2 and p27 in breast cancer and indicate its signicance and corrlativity with each other .Method We selected 80 cases of surgical specimens of breast cancer. The expression of Skp2 and p27 in breast cancer were studied by using tissue chip technique and S- P immunohistochemical method. Result In 80 cases of breast cancer, the positive rates of Skp2 and p27 were 46. 3%、 41. 3% respectively, The Skp2 positive rate in cases of p27 positive was 15. 2% and in cases of p27 negative was 68. 1%, there being significant difference between the two groups(P<0. 01). Conclusion In breast cancer, the expression of Skp2 and p27 was not related to the histological type. High expression of Skp2, low expression of p27 were closely related to the extent of invasivation and lymph node status. The high expression of Skp2 was obviously negatively related to the low expression of p27. In the cell cycle regulation of breast cancer, p27 is a negative factor and can inhibit cell proliferation , and Skp2 can promote p27 to decline so that it can improve cell proliferation. The large-scale application of tissue chip is viable to test the clinical specimens efficiently. It shows that the method is save-timed, convenient, economy and accurate.
