苜蓿HD-Zip转录因子基因Mfhb-1的生物学功能初探

【摘要】 以苜蓿转化植株为材料,利用苜蓿的直接体细胞胚胎发生体系,对苜蓿 Mfhb-1转录因子基因的生物学功能进行了研究。结果表明:正义转化植株 F4(转入基因编码区序列)产生体细胞胚胎时期较47/1-5对照、反义转化植株 D3(转入编码区的反向序列)和 C5(转入编码区+uORF)早;正义转化植株 F4形成的体细胞胚胎数量远高于47/1-5(对照),D3、C5植株产生的体细胞数量均较47/1-5(对照)少,但 C5植株产生的体细胞数量较 D3稍多。从以上结果可以推测出属于 HD-Zip 转录因子的 Mfhb-1基因能够促进苜蓿体细胞胚胎的发育,并能够提高苜蓿体细胞胚胎发生的数量。
【Abstract】 Sense and anti-sense transgenic alfalfa plants (F4 and D3) were used to investigate the biological func- tions of the HD-Zip transcription factor gene Mfhb-1 via direct somatic embryogenesis system in alfalfa.The over-expression of the Mfhb-1 in sense transgenic plant F4 generated a much higher number of somatic embbryos,while its lower-experesion in D3 produced less number of embryos,compared with that in control plant 47/1-5.The expression of full length of the Mf- hb-1(CDS region of Mfhb-1 plus an upstream uORF region) in the transgenic plant C5 yielded the number of embryos less than that in control plant,but slightly higher than that in D3.All the results indicated that the expression of the Mfhb-1 may closely associate with somatic embryogenesis,and possibly has the function to enhance somatic embryo development and somatic embryogenesis in alfalfa.
