转双价抗虫基因水稻的获得

【摘要】 采用根癌农杆菌介导法将2个抗虫基因(sbk 和 sck)同时转入3个水稻品种中,通过一系列的分子检测(PCR 和 RT-PCR)分析,结果表明:这2个基因已经成功转入3个水稻品种中并得到了表达;同时还获得1株不含抗生素筛选标记基因(hpt)和抗虫 sbk 基因,却含抗虫 sck 基因的转基因植株,命名为 YJ27-5sck。用该植株作为供体,通过杂交将 sck 抗虫基因转育到另外5个水稻品种中且同样得到了表达,获得了不含筛选标记基因的转 sck 基因阳性植株。

【Abstract】 Two insecticidal genes (sbk and sck) were transferred into three rice varieties mediated by agrobacterium tumerfaciens approach,the results of PCR identifications indicated that the two genes had been integrated into the genomes of three rice varieties and the targeted genes were successfully expressed.And a transgenic individual whose antibiotics se- lection marker gene (hpt) and sbk insecticidal gene were negative but the sck insecticidal gene was positive was distinguish- ed and was named as YJ27-5sck.The insecticidal gene sck was successfully transferred into other five rice Uarieties using YJ27-5sck as donor,and sck transgenic rice plants were obtained.
