Bt+CpTI抗虫基因转化大豆的研究

【摘要】 本试验以大豆子叶节为外植体,以农杆菌菌株EHA101为转化菌,研究受体基因型、农杆菌侵染时间、预培养时间及培养添加物对大豆转化效率的影响并用Bt+CpTI基因转化大豆。结果表明,不同基因型、不同侵染时间对大豆子叶节的转化率有较大影响,浸染26 min时具有最高的表达率;预培养3天、在共培养的过程中添加2mg/L的脯氨酸或硝酸银对大豆的转化效率都有提高;Bt+CpTI基因的导入能够提高受体抗大豆食心虫的能力。
【Abstract】 This experiment was conducted to transfer a insect resistant gene (Bt + CpTI) into soybean and optimize the Agrobacterium mediated transformation in cotyledonary nodes. The results indicated that both recipient genotypes and infection times affect the transformation frequency of Agrobacterium mediated in soybean cotyledonary nodes. Infecting for 26 minutes is better for soybean transformation in cotyledonary nodes. Adding AgNO3 and pro-line in co - cultivation increased the transformation frequency and the concentration of 2mg/ L proline and 2mg/L AgNO3 was optimal. Three days pre-cultivation for explants were good for transformation in cotyledonary nodes. The ability of transgenic plants resistant to soybean borer was increased.
