高赖氨酸蛋白基因SBgLR的克隆及其对提高玉米种子中蛋白质和赖氨酸的作用

【摘要】 从马铃薯(Solanum tuberosum L.)中克隆的SBgLR基因由3个外显子和2个内含子组成,编码211个氨基酸,其赖氨酸重量比为18.93%,是一个天然存在的高赖氨酸蛋白基因。构建了两个含SBgLR基因组DNA的植物表达载体,用基因枪转化方法将其导入玉米(Zea mays L.)的胚性愈伤组织,经PCR、点杂交和Southern blot检测,表明外源基因已整合到玉米的基因组中,并稳定遗传至下一代。通过检测R1转基因玉米种子的蛋白质和赖氨酸含量发现,96.7%的植株蛋白质的含量都提高,提高幅度为5.4%-58.1%;93.3%的植株赖氨酸含量也有提高,提高幅度为3.2%-54.8%。R2植株种子中97.2%的植株蛋白质和赖氨酸含量有提高,提高幅度分别为19.2%-55.1%和3.2%-51.6%,得到6个蛋白质和赖氨酸均提高30%以上的株系。实验结果表明SBgLR基因可以提高玉米种子中的蛋白质和赖氨酸含量。
【Abstract】 A SBgLR gene contained three exons and two introns was isolated from potato (Solanum tuberosum L. ) and encoded a protein of 211 amino acids. It was a natural lysine-rich gene which lysine composition was 18.93% (W/W). Plant expression vectors p19ZKgH (seed-specific promoter 19Z:: SBgLR genomic DNA) and pAkgH (Adh1 promoter: SBgLR genomic DNA) were transformed into maize(Zea mays L.) embryonic callus by microprojectile bombardment. The transformation maize plants were analyzed by PCR, Dot blotting and Southern blotting to verify the integration of SBgLR gene into maize genome. The contents of crude protein and lysine in mature seeds of R1 transgenic plants were analyzed respectively and found that protein content of 96.7% transgenic plants had an increase of from 5.4% to 58.1 % and lysine content of 93.3% transgenic plants had an increase of from 3.2% to 54.8 %. In R2 transgenic plants, the crude protein and the lysine contents of 97.2 % transgenic plants were increased from 19.2% to 55.1 % and from 3.2% to 51.6% respectively. Six transgenic lines with the increases of 30% in both protein and lysine contents can be used further breeding. The result showed SBgLR gene could be used to improve the protein and lysine contents of transgenic maize seed.
