人新的蛋白激酶抑制剂HBRP功能的研究

【摘要】 目的:研究新基因HBRP(Human BSP-Related Protein)的生物学功能。我们从人精子中纯化了一个PKC(protein kinase C)的抑制剂,但经测序发现与BSP(bovine seminal plasma)序列完全相同,进一步的研究发现BSP不仅能抑制PKC的活性,还能抑制酪氨酸蛋白激酶的活性。而在人的生殖系统中与BSP蛋白功能相关的蛋白未见报道。我们克隆了人与BSP蛋白相关基因的cDNA序列,将其命名为HBRP,并在GenBank注册,登录号为AF27914。已证明该基因在人肾、肝、脾、肠、胃、胰腺、骨骼肌、心肌、脑、肺等11中组织中均有阳性表达。方法:利用真核重组表达质粒pcDNA3/HBRP转染人胚肾HEK293细胞,建立了稳定的真核表达细胞系,并用放射自显影检测该细胞系的PKC活性。结果:HBRP蛋白对PKC活性有明显的抑制作用,PKC活性明显降低。结论:确定了人新的精子结合蛋白HBRP的功能之一是对PKC活性有抑制作用。
【Abstract】 Objective: To study the biology function of novel HBRP(Human BSP-Related Protein) gene.We purified an inhibitor of PKC(protein kinase C) from human sperm,but we found that the sequence of the inhibitor is simillar with BSP(bovine seminal plasma) entirely.The further study indicated that BSP inhibite not only the activity of PKC,but also the activity of tyrosine protein kinase.There is no report about related protein to BSP in human genital system.We cloned cDNA sequence of related protein to BSP in human, named HBRP, and registen in GenBank(AF27914).We have proved that the gene is widely expressed in these tissue in the human organ such as testis、kidney、intestines、stomacu、lung、 muscle、liver、heart and spleen. Mthods: we transfected HEK293 cell by recombination vector pcDNA3/HBRP,constructed stable expression cellular line. We detected the activity of PKC in the transfected cellline by radioautography. Results: HBRP protein inhibit the activity of PKC obviously. Conclusion: One of the new function of novel sperm binding protein in human is to inhibit the activity of PKC.
