新的蛋白激酶抑制剂HBRP基因真核表达体系的建立

【摘要】 目的:建立一个新基因HBRP(Human BSP-Related Protein)的稳定真核表达细胞系,对该基因功能进行研究。在研究牛精浆蛋白(bovine seminalplasma,BSP)及其相关蛋白在受精卵发育过程中的作用时,我们在人睾丸组织中发现并克隆了一个与BSP蛋白相关新基因的序列,其开放阅读框架(open readingframe,ORF)编码了一个含223个氨基酸残基的蛋白质,氨基酸序列中含有四个纤连蛋白Ⅱ型结构域,预测该蛋白是与BSP蛋白功能相关的结合蛋白。方法:为进一步研究该新基因的生物学功能,我们利用重组质粒pEGFP-C2/HBRP转染人胚肾HEK293细胞,通过G418筛选出药物抗性克隆。结果表明,HBRP蛋白对PKC活性有明显的抑制作用。结论:HBRP蛋白可能一个是蛋白激酶新的抑制剂。
【Abstract】 Objective: We constructed stable eukaryotic expression cell line to further study the biological function of the novel HBRP(Human BSP-Related Protein) protein.The novel gene HBRP which is related to proteins of bovine seminal plasma (BSP) in human testis was found and cloned when we studied the important roles of BSP in fertilization and the development of zygote. Its open reading frame(ORF) encoded a protein of 223 amino acids. There were 4 fibronectin Ⅱ-domains in the amino acid sequence. It is estimated that HBRP is a kind of binding protein whose function is similar to that of BSP. Methods: to further study the biological function of HBRP,we transfected HEK293 cells by pEGFP-C2/HBRP recombination plasmid, and positive colony of the gene is selected. The results showed that HBRP protein inhibited the activity of PKC obviously. Conclution: HBRP protein maybe a new inhibitor of PKC.
