131I-chTNT在荷人结肠癌裸鼠动物模型中的放射免疫显像

【摘要】 目的评价131I-单克隆抗体chTNT与结肠肿瘤的亲和性,以了解其用于结肠癌定位诊断和导向治疗的可能性。方法18只荷人结肠癌裸鼠动物模型腹腔注射5.55MBq131I-chTNT后1、2、3、5、7天,分别作放射免疫显像,并逐日分组杀死,行组织分布测定,并与131I对照组比较。结果实验组注射标记抗体后1天肿瘤组织清晰显影,并持续显示至实验结束。瘤体内放射性维持相对稳定水平,131I-chTNT在结肠癌组织内的有效半减期为6.8±1.8天。其他脏器组织放射性浓聚少,并逐渐消失。第7天T/NT比值达14.8±5.3。结论13I-chT-NT具有导向结肠肿瘤的特性,为用于结肠癌放射免疫显像和导向治疗提供了依据。

【Abstract】 Purpose In order to evaluate the targeting activity in the animal model with human colon cancer , the 131I - chTNT radioimmunoimaging was explored. Methods Radioimmunoimages were taken on different intervals after injection of 131I - chTNT 5.55MBq to the nude mice, and tissue distribution was measured. The results of 131I - chTNT monoclonal antibody group were compared with that of 131I group. Results The experimental group developed tumor positive images after one day of radio - labeled monoclonal antibodies injection and held on until the end of the experiment. The radioactivity in tumor mass was stable, and the half life of 131I-chTNT in colon cancer mass was 6.8 ± 1.8 days. There was no special radioactivity accumulation in nonnal liver tissue in the nude mice and the radioactivity in it disappeared rapidly. Statistic indicated the tumor colon ratio in 7 days were 14.8. Conclusions: Our results suggest that 131I - chTNT monoclonal antibody has a considerable targeting activity, and provide an evidence for that it can be used as a new radiopharmaceutical agent for the imaging and radio therapy of colon cancer.
