针灸调节免疫抑制效应的机制研究

【摘要】 目的从免疫细胞因子在中枢神经系统的基因和蛋白表达水平,免疫靶细胞和外周血血清中细胞因子含量的分子水平变化,探索内源性细胞因子在针灸调节免疫抑制效应中的调控机制。研究方法和结果采用BALB/C小鼠H22移植性实体瘤造成免疫抑制模型,随机分为荷瘤组、针灸治疗组、非穴位治疗组以及正常组和正常小鼠治疗组,共五组。1.RT-PCR、原位杂交、免疫组织化学ABC办法,结果显示针灸信号引起中枢神经系统大脑皮层神经细胞胞浆IL-1β、IL-2、IL-6 mRNA及蛋白表达改变;2.艾灸刺激导致脾淋巴细胞IL-2及其受体IL-2Rα、IL-2Rβ、IL-2RγmRNA 表达变化,125I-IL-2放射配基检测脾淋巴细胞IL-2受体(IL-2R)数量改变;IL-2R介导的Jak-Stat 信号途径中Jak1、Jak3、Stat5a和Stat5b mRNA表达变化;3.艾灸刺激后的外周血清能明显提高脾淋巴细胞细胞因子IL-1β、IL-2水平;4.SDS-PAGE电泳初步发现,荷瘤小鼠经艾灸治疗后外周血清在26.5KD表达变化。结论艾灸信号引起大脑皮层IL-1、IL-2、IL-6 mRNA改变,导致相应与免疫抑制效应有关的特异蛋白表达,作用于免疫靶细胞引起相关细胞因子表达水平改变, 特别是肿瘤免疫最重要的调节因子IL-2和IL-2Rα、β、γ各亚基以及IL-2R介导的Jak-Stat信号转导途径中Jak1,3、Stat5a、5b表达变化,从而调整免疫网络,正向调节肿瘤免疫抑制状态。

【Abstract】 Objective: To investigate the molecular mechanism of moxibustion on modulating immunosuppression induced by H22 tumor cell, through endogenous cytokines IL-1, IL-2, IL-6 from gene transcription to protein expression in cerebral cortex and spleen lymphocyte, and cytokine levels in peripheral blood serum of tumor-bearing mice. Methods and results: Immunosuppressive B ALB/C mice transplanted H22 tumor cell, were randomly divided into 5 groups: the Tumor control group, the Moxibustion treatment group, Moxibustion treatment on non-acupoint group, the Normal control group, and the Moxibustion on normal mice group. 1. Results by RT-PCR, ISH and Immunohistochemistry ABC methods showed: the expression of IL-1, IL-2 and IL-6 mRNA and protein was responded to moxibustion treatment in cerebral cortex; 2. the expression of IL-2 and its IL-2Rα、IL-2Rβ、IL-2Rγ mRNA was responded to moxibustion treatment; the number of IL-2 receptor in spleen lymphocyte was increased after moxibustion treatment by 125I-IL-2 Radio-binding Assay; It also showed that the expression of Jak1, 3 and Stat5a, 5b mRNA in Jak-Stat pathway of IL-2R signal transduction was upregulated in tumor-bearing mice after moxibustion treatment. 3. the level of IL-1βand IL-2 produced by the spleen lymphocyte was increased in the induction of peripheral blood serum after moxibustion treatment on tumor-bearing mice. 4. The results also showed 26.5KD of electrophoresis strips was reappeared after moxibustion treatment in tumor-bearing mice by SDS-PAGE, compared to no this electrophoresis strips in tumor control group. Conclusion: The respond that the expression of endogenous cytokines IL-1, IL-2,IL-6 mRNA in cerebral cortex was induced by moxibustion signal, results in the expression changes of relative cytokines in immune effector cell, especially the expression changes of IL-2 production, IL-2R subunit IL-2Rα、β、γ, and the expression changes of Jakl, 3、Stat5α、5b in Jak-Stat signal transcondution pathway. Acupuncture/moxibustion could upregulate immunosuppressive state in tumor-bearing mice by the modulation of neruo- endocrine- immune network.
