四神方对TNBS诱导的小鼠结肠炎的作用及免疫调节机制

【摘要】 目的:观察四神方对TNBS诱导的小鼠实验性结肠炎的防治作用及免疫调节机制。方法:小鼠麻醉后直肠内灌注含1.5mg的TNBS的50%乙醇溶液制备结肠炎模型。2d后开始四神方灌胃给药,连续8天。观察小鼠的一股状态和DAI评分、结肠组织学及结肠中MPO变化,并通过细胞因子的分泌、T淋巴细胞增殖及相关协同刺激分子的表达探讨其免疫调节机制。结果:四神方对TNBS结肠炎小鼠的一般状况及DAI评分有改善作用、并能缓解结肠局部的炎症,减轻组织中炎细胞的浸润。四神方可降低结肠局部IL-12/23 p40和IFN-γ的水平、抑制肠系膜淋巴结中T 淋巴细胞协同刺激分子的表达及其增殖。结论:四神方能通过显著的抑制T淋巴细胞活化达到防治TNBS所诱导的小鼠结肠炎的作用。

【Abstract】 Objective: To explore effect and mechanism of immunoregulation of Sishen recipe treats experimental colitis induced by trinitrobenzene sulfonic acid in mice. Methods: Mice colitis was induced by rectal administration of 1.5mg trinitrobenzene sulfonic acid (TNBS) in 50% ethanol. After 2 day mice were treated with Sishen recipe daily. Changes in disease activity index (DAI) together with histolpgic scores were evaluated, and myeloperoxidase (MPO) in colonic tissue was detected. Cytokine production was assessed by ELISA. T lymphocyte proliferation was evaluated using MTT method. Costimulatory molecule expression was analyzed by FACS. Results: Sishen recipe therapy significantly reduced DAI and histological scores in mice, and alleviate inflammation in colonic tissue. Sishen recipe not only decreased significantly cytokine IL-12/23 and IFN-γ level, but also inhibited T lymphocyte proliferation activation and expression of costimulatory molecule. Conclusions: Therapeutic efficacy of Sishen recipe on mice colitis may rely on its action of suppressing T lymphocyte activation significantly.
