【摘要】 目的:观察生姜泻心汤对伊立替康(Irinotecan,CPT-11)所致迟发性腹泻模型大鼠肠黏膜免疫系统的影响,探讨其对迟发性腹泻的预防作用。方法:健康SD大鼠18只,随机分为正常对照组,腹泻模型组,中药+腹泻模型组,尾静脉注射CPT-11 150mg／kg／day,连续两天,复制迟发性腹泻模型;中药+腹泻模型组于注射CPT-11前3天开始,每天一次用生姜泻心汤灌胃,共用 10天。对各组大鼠腹泻情况进行评分;光镜下根据chiu肠黏膜损伤分级法对大鼠肠黏膜损伤程度进行分级;免疫组织化学染色法标记大鼠肠黏膜CD4+、CD8+T淋巴细胞及SIgA,用图像分析仪统计计算其含量。结果:中药+腹泻模型组大鼠迟发性腹泻发生率低于腹泻模型组(P<0．01);光镜下对肠粘膜损伤分级亦较模型组轻微(P<0．05);肠黏膜上CD4+、CD8+T淋巴细胞及SIgA表达亦增多(P<0．05)。结论:生姜泻心汤可提高大鼠肠黏膜免疫功能,预防迟发性腹泻的发生。更多还原

【Abstract】 Objectives: To observe the effect of Zingiberis Recentis Decoction for Purging Stomach-Fire on intestinal mucosal immune system of rats with delayed-onset diarrhea caused by Irinotecan, to study the prevention of Zingiberis Recentis Decoction for Purging Stomach-Fire on delayed-onset diarrhea caused by Irinotecan. Methods: Sprague Dawley male rats(n=18) , randomly assigned to three groups ,normal group , delayed-onset diarrhea group , Herb group,injected with 150 mg/kg body weight/day CPT-11 for 2 consecutive days in the tail vein , to establish a model of delayed-onset diarrhea; Herb group were treated with Zingiberis Recentis Decoction three days before CPT-11 injected, once a day. Scoring of delayed-onset diarrhea. CD4+、CD8+ T-lymphocytes and SIgA in rats’ intestinal tissues (ileum, cecum and colon) were labeled by immunohistochemical staining and there number was calculated using imaging analyzer. Results: The incidence of diarrhea in herb group was lower than delayed-onset group(P<0.05 ); Compared with diarrhea-onset group, numbers of CD4+、CD8+ T-lymphocytes and SIgA in rats’ intestinal mucosal immune system increased significantly in herb group(P<0.05). Conclusion: Zingiberis Recentis Decoction for Purging Stomach-Fire could enhance the immune function in intestinal mucosa on rat with CPT-11 induced diarrhea-onset, and it could prevent the delayed-onset diarrhea caused by CPT-11.更多还原
