脂多糖刺激前后小鼠肺肝脾组织中TLRs等受体的基因表达情况

【摘要】 目的:了解LPS对肺肝脾组织中与LPS信号转导有关的受体的基因表达情况的影响。方法:LPS刺激前后取小鼠肺、肝、脾组织总RNA,RT-PCR半定量测定LBP、CD14、TLR2、TLR4 和TN F-α等基因的表达情况。结果:正常组织中,肺有TLR2的表达,肝有TLR2、CD14、LBP 的表达,脾有TLR2的表达;注射LPS后的5小时内,肺组织TLR2、TLR4、CD14和TNF-α,肝组织TLR2、CD14、LPB和TNF-α,脾组织TLR2、TLR4、CD14和TNF-α的基因表达呈逐渐增加的趋势。结论:正常情况下TLR2等基因在不同组织中的表达有差异,LPS刺激5小时内提高各基因的表达,从而提高机体对LPS等病原体模式识别分子的反应性。

【Abstract】 Objective: to investigate the gene expression of some LPS receptors after LPS stimulation and the effect of Emodin on these genes. Methods: the total RNA from normal and LPS-challenged mice was extracted by Trizol reagent and the gene expression of TLR2, TLR4, CD14, LBP and TNF-α was measured by semi-quantitative RT-PCR; in the emodin groups, mice were pretreated by emodin for 30min. Results: the gene of TLR2 was expressed in normal lung and spleen tissue, and TLR2, CD14, LBP in liver. After challenged by LPS, the expressions of TLR2, TLR4, and TNF-αin lung, TLR2, CD14, LBP and TNF-αin liver, TLR2, TLR4, CD14, and TNF-αin spleen were increased at 1, 3, and 5 hrs. Emodin presented inhibiting effects on the expression of these genes in a vary extent. Conclusion: LPS alerted the ability against pathogen-associated molecule by inducing or enhancing the expression of genes that involved in the LPS signal transduction; while the emodin may blunted the reaction by inhibiting the gene expression.
